Would this work for a 2019 21.5 4K iMac?

No. The tape strips work for a 27" monitor -- the length and placement of the notches won't suit a 21.5"

How difficult is it to remove this adhesive if the machine needs to be re-opened once sealed back up with these strips?

It is not hard to remove at all. One week after i used those strips, the screen fell off of my computer while i was sleeping. I bought a new screen but the logic board got damaged so now i have an 2020 i9 iMac that does not work. I would recommend the the 3M strips instead.

Is there any diagram how to use it? The strips look confusing and fragile. There must be a diagram provided either in this product page or in the box.

If you're talking about where the strips need to be installed I had wondered the same thing until I searched also on Ebay then it showed a diagram. #11 goes in the top left, #12 top right, #13 Right side and so forth all going clock wise in # order.
