How many medium sized pills would you say fit in each compartment?

3 or 4. it's hard to say because two of the compartments are bigger than two others. I take about 9-12 vitamins a day and I can fit them in there.

are there instruction to wash before first use like the drug store pill organizers. I never washed mine

No instructions provided. Was not an original Pill Pro. A knockoff arrived in the original box, the days printed on it were smudged off in areas. Cheap.

Are replacement daily compartments available? I lost Wednesday on a trip, and one of the latches on Saturday is broken.

Unfortunately, I do not have any replacement compartments.

Do the lids fit tightly enough to use for a powdered supplement?

Yes, the lids are tight! In each day of the week Their are 4 sections. Not sure how much powder you need in each but it would definitively work for you! I love that I took a chance and purchased this! I love it!
