Yes, you should use speaker wire. Think of the Buttkickers as a silent subwoofer. They vibrate in synch with the subwoofer channel and supplement your subwoofer(s). You will need an amp to power the Buttkickers also. You usually need a dedicated amp.

How do I hook it up to a surround sound 5.1 system?

You need an Amplifier. ButtKicker sales their own brand or you can buy a third party brand.

I want to mount this to an office chair. How many transducers will I need? 1 or 2?

If you are just mounting it to an office chair you will only need 1.
