Does the box come with any labeling tape (to get started)?

Yes, it does: one roll of 1/2" x 10' , black ink on white, laminated (glossy). It also comes in 2 other sizes - 1/4" and 3/8" (not in the same box, sold separately).

Can you print multi line labels? and if so how many lines

It prints up to two lines on a 1/2" and 3/8" tape. Although there are six text sizes available, the text sizes depend on the label width you have selected. The 1/2" tape can handle 8, 10 and 12 point text size; the 3/8" only handles 8 and 10 point text size. I hope I didn't confuse you. You can print the "User Guide" from the DYMO website.

Can you print directly from a computer using this?

It hooks up to computers and you have to download the software but then you can type up labels on your computer and they will print directly from this machine. I had a project where I had to make 500+ labels and I typed them all on the computer and this labeler printed them all like a champ. Any font any style!

Can you switch the cartridges in and out, or do you have to finish each roll before changing?

No problem changing cartridges at any time. In fact I have not had any issues or problems with this unit at all.
